- In-gel digestion
- from one or two dimensional gels spots are excised by a robotic device to avoid cheratins contamination. After trypsin digestion peptides are analysed by mass spectrometry.
- In liquid digestion
- trypsin digestion performed in a liquid solution. The buffer should be compatible with the enzyme activity.
- Automatic digestion
- digestion with trypsin (or other proteolitic enzymes) by Liquid handler really convenient for whole proteome identification.
- Infusion analysis of proteins or peptides
- it requires at least 50fmol/µl and 5µ of protein solution.
- Zip Tip C18purification
- it is mandatory if the sample contains contaminants such as salt.
- MS/MS analysis
- in order to obtain information on the aminoacid sequences of peptides we use a QqTOF (Q-Star) to fragment the precursor ions. This tecnique is very powerful to identify proteins from non sequenced organisms and to correctly identify post translational modifications on specific aminoacids.
- Databases search
- databases searches are performed by Mascot distiller to identify proteins from PMF and MS/MS and for protein quantitation after isotope labelling (ICPL, iCAT, iTRAQ).
